Abstract: 

BACKGROUND LEISHMANIASIS IS ENDEMIC IN 5 CONTINENTS AND 98 COUNTRIES AROUND THE WORLD AND THREATENS 350 MILLION PEOPLE WORLDWIDE ANNUALLY. SAMPLING AND DIAGNOSIS USING DIRECT SMEAR IS COMMON IN ENDEMIC AREAS, BUT THIS METHOD HAS LOW SENSITIVITY DESPITE THE HIGH SPECIFICITY. ALSO DIFFERENT SPECIES OF PARASITE DO NOT GROW EASILY IN CULTURE MEDIUM AND MAY TAKE WEEKS TO GROW. WE AIMED TO EVALUATE THE RESULTS OF DIRECT SMEAR AND SKIN BIOPSY OF PATIENTS SUSPECTED TO CUTANEOUS LEISHMANIASIS USING PCR-RFLP AND COMPARISON OF ITS SPECIFICITY AND SENSITIVITY WITH CONVENTIONAL METHOD.MATERIAL AND METHODS SKIN BIOPSIES FROM 100 PATIENTS REFERRED TO HEALTH CENTERS IN ISFAHAN PROVINCE WITH CUTANEOUS ULCERS SUGGESTIVE OF LEISHMANIASIS WERE OBTAINED INTO MICRO-TUBES CONTAINING 70% ETHANOL. SMEARS WERE PREPARED FROM THE EXUDATES AND STAINED WITH GIMSA.
