REPEATED ADMINISTRATION OF MORPHINE ALTERS HISTAMINE- INDUCED ANXIOGENIC EFFECT PRODUCED BY INTRA-VENTRAL HIPPOCAMPAL MICROINJECTION

Abstract

 It has been suggested that HISTAMINE have modulatory influence on ANXIETY-RELATED BEHAVIORs both in animals and humans. In the present study, the effects of intra- VHC injections of HISTAMINE and pyrilamine (HISTAMINE H1 receptor antagonist) on ANXIETY-RELATED BEHAVIOR have been investigated in MORPHINE-sensitized and naïve rats. Rats were bilaterally cannulated in the VHC by a stereotaxic instrument. The elevated plus-maze which is a useful test to investigate the effects of anxiogenic or anxiolytic drugs in rodents was used as a model of anxiety. Intra-VHC HISTAMINEinduced anxiogenic effect was reversed in animals that had previously received the 3- days MORPHINE (7.5 mg/kg) followed by 5 days free of the drug. Furthermore, intra-VHC administration of pyrilamine (5, 10 and 20 μg/rat) decreased %OAT (Open Arm Times) and %OAE (Open Arm Entries) indicating an anxiogenic response. Intra-VHC pyrilamine- induced anxiogenic effect didn’t change in animals that had previously received the 3-days MORPHINE (7.5 mg/kg) followed by 5 days free of the drug. Our results indicated that while HISTAMINE induces anxiogenic response in rats, this behavior was attenuated in MORPHINE sensitized rats. The present data also show that microinjection of pyrilamine into the VHC resulted an anxiogenic response. On the other hand, our results indicated that intra- VHC pyrilamine- induced anxiogenic effect has didn’t change in MORPHINE sensitized rats. Therefore, the effect of repeated administration of MORPHINE differentiates between HISTAMINE and its antagonist. No significant change in the LOCOMOTOR ACTIVITY was observed following the drug treatments in this experiment.
