COMPARISON OF EFFECTS OF GONADECTOMY ON DEMYELINATION AND REMYELINATION PROCESSES IN OPTIC NERVES AND CHIASM OF ADULT MALE RATS

Abstract

 Multiple sclerosis is a neurodegenerative disease with a higher incidence in adult women than men. Myelin sheaths are destroyed and demyelinated in the nervous system and some sensory disorders and paralyses are developed. The spontaneous REMYELINATION occurs after DEMYELINATION. Sex hormones like TESTOSTERONE have protective effects on development and growth of nervous system. In this study, we studied the effect of GONADECTOMY on DEMYELINATION and REMYELINATION processes by using Visual EvokedPotential (VEP) recording in adult male rats. We injected 2μl of LPC 1% into the chiasm of male Spruge Dawley rats using stereotaxic instrument under anesthesia. Then, we recorded VEP signal in both gonadectomized and control rats at 2, 7, 14 and 28 days post-injection. The latencies of P and N waves were measured and compared. Following LPC injection, the latency of both waves was increased in control and gonadctomized rats. The VEP latency was more increased on 7 and 14 days post-lesion in gonadectomized group and the latency was more decreased in gonadectomized rats on day 28 post-lesion. Our results showed that GONADECTOMY increases DEMYELINATION and decreases REMYELINATION in optic nerves and chiasm of adult male rats. It seems that TESTOSTERONE can protect myelin sheath and increase its regeneration.
